چکیده

Abstract Background The widespread popularity of porcine circovirus type 2(PCV2) has seriously affected the healthy development pig industry and caused huge economic losses worldwide. A rapid reliable method is required for epidemiological investigation evaluating effect immunization. However, current methods PCV2 antibody detection are time-consuming or very expensive rarely meet requirements clinical application. we have constructed platform expressing nanobody(Nb)?horseradish peroxidase(HRP) fusion protein as an ultrasensitive probe to detect antibodies against Newcastle disease virus(NDV), previously. In present work, Nb-HRP protein-based competitive ELISA(cELISA) simple was developed using this anti-PCV2 in serum. Results Using phage display technology, 19 anti-PCV2-Cap nanobodies were screened from a PCV2-Cap immunized Bactrian camel . With platform, PCV2-Nb15?HRP then produced used sensitive reagent developing cELISA anti?PCV2 antibodies. cut?off value 20.72 %. Three hundreds sixty serum samples tested by both newly commercial kits. sensitivity specificity 99.68 % 95.92 %, respectively. coincidence rate two 99.17 When detecting 620 samples, good consistent (kappa = 0.954) found between results those Conclusions brief, based rapid, low-cost, useful nanobody-based tool serological evaluation vaccine efficacy indirect diagnosis infection.

Development of Isothermal Recombinase Polymerase Amplification Assay for Rapid Detection of Porcine Circovirus Type 2

Porcine circovirus virus type II (PCV2) is the etiology of postweaning multisystemic wasting syndrome (PMWS), porcine dermatitis, nephropathy syndrome (PDNS), and necrotizing pneumonia. Rapid diagnosis tool for detection of PCV2 plays an important role in the disease control and eradication program. Recombinase polymerase amplification (RPA) assays using a real-time fluorescent detection (PCV2 ...

Rapid Detection of Porcine Circovirus Type 2 by TaqMan-based Real-time Polymerase Chain Reaction Assays

PCV2 is the primary causative agent of porcine circovirus-associated disease (PCVAD). In this study, a TaqMan-based real-time polymerase chain reaction (PCR) assay targeting ORF2 gene of PCV2 was developed and their sensitivities and specificities were investigated. The results indicated that the standard curve had a wide dynamic range (102-107copies/μL) with a linear correlation (R2) of 0.997 ...

Immunology of porcine circovirus type 2 (PCV2).

The emergence of porcine circovirus type 2 (PCV2) associated diseases and particularly postweaning multisystemic wasting syndrome (PMWS) was a shock for the swine industry and formulated a considerable challenge for researchers in the area of viral immunology in swine. The unique features of PMWS of which emaciation and lymphoid depletion were the most evident indicated a deep involvement of th...

Coincidental detection of genomes of porcine parvoviruses and porcine circovirus type 2 infecting pigs in Japan

The infection status of 15 viruses in 120 pigs aged about 6 months was investigated based on tonsil specimens collected from a slaughterhouse. Only 5 species of porcine parvoviruses and porcine circovirus type 2 (PCV2) were detected at high frequencies; 67% for porcine parvovirus (PPV) (PPV-Kr or -NADL2 as the new abbreviation), 58% for PPV2 (CnP-PARV4), 39% for PPV3 (P-PARV4), 33% for PPV4 (PP...
